Description:
My doctoral research focused on the initial transcriptional defence responses of sunflower against Sclerotinia head rot (SHR), caused by the fungus Sclerotinia sclerotiorum. In order to evaluate the level of resistance in various inbred lines, I used phenotypic, genotypic, and molecular analyses.
The Purity of the materials was ensured through SSR molecular markers, and the resistance levels were evaluated using incidence, severity, intensity, and the area under the disease-progress curve (AUDPC). To analyze the changes in gene expression, I used microarrays and identified response genes based on factors such as fold changes, p-values, co-expressed genes, and multivariate analysis. To achieve this I employed various tools, including Infostat, MapMan, R + WGCNA library, Blast2Go, MeV, and Galaxy.
The identified cluster of differentially expressed genes were specific to the susceptible or resistant lines, indicating that the responses were independent. Their functional annotation identified various functions, including proteolysis, membrane receptors and Ca2+ signaling, cellular redox, and plant hormones auxins and ethylene
Morover, my research identified 43 candidate resistance genes (and 47 putative susceptible genes) with crucial roles in plant defense, such as: oxidative stress detection, transcription factors, ubiquitination, peptidases inhibitors, membrane zinc transporter proteins, synthesis of amino acids, and dynamics of the cytoskeleton. I also designed two techniques to detect the early presence of S. sclerotiorum, prior to the onset of the first SHR symptoms appear. One approach relied on optical observations while the other on PCR.
My work has deepened our understanding of the complex molecular mechanisms involved in the triggering and evolution of S. sclerotiorum infectious process in sunflower as well as to the identification of genes involved in different stages of SHR disease, and has the potential to enhance the improvement of this oleaginous crop.
